This knowledge is needed to provide new strategies that will enhance the capabilities of engineering tissue implants. This proposal aims to determine how the actin cytoskeleton regulates both protrusion formation and the mechanical properties of cell substrates. Specifically, the goal of this proposal is to determine how the F-actin structures that support protrusion formation are regulated by contractile force. The hypothesis is that the initial sites of F-actin protrusion formation are sites where contractile force is generated and that this regulation occurs through release of the RhoGTPase Cdc42 from its GDP bound form. The mechanosensory mechanism is hypothesized to be actin polymerization through sites of active RhoGTPase signaling. The focus of this proposal is on the following specific aims: Aim 1, to test the hypothesis that actin polymerization is necessary for the generation of contractile force; Aim 2, to test the hypothesis that RhoGTPase signaling at nascent protrusions regulates the mechanics of actin protrusion formation; and Aim 3, to test the hypothesis that contractile force generated by the actomyosin cytoskeleton is essential for cell protrusion formation. The experiments in this proposal will use an interdisciplinary approach that integrates live cell imaging, fluorescent, and atomic force microscopy to investigate the microscale forces of cell protrusion formation. The research will be done in the laboratory of co-Principal Investigator Michael Davidson, the Director of the Biointerfaces Institute.
